Isolation and characterization of mesophilic protease from marine environment

It is important to explore marine resources by finding a protease that can be applied to food industry. Indonesia is one of the mega-biodiversed countries, the chance to find potential enzyme is very great. Sample of sea water obtained from Bali's beach area (Gondol) was taken to be grown in a skim medium. Growing bacteria are then isolated and purified. The colony with the best clear zone production was isolated and produced in liquid skim medium (2%). Growth curve and optimization of production time were conducted before enzyme's production. The enzyme was concentrated using ultra filtration having MWCO of 10000 Da. Characterization was conducted including optimum temperature, optimum pH, heat stability and effects against metal ions. Three protease producing isolates were obtained (white, white irregular and yellow). White irregular or later called as WI isolate was taken into further research as it produced the best clear zone. Protease produced by WI isolate worked best at 37┬░C and at pH 5. It was stable at 37┬░C and the activity could be increased with addition of Zinc and Calcium ion as chloride salt (1mM), and decreased by EDTA, Cobalt, Manganese and Iron. WI isolate produce maximum protease at 12 hours. With the enzyme's stability at 37┬░C, it can be applied in the food industry which runs a process at around 30┬░C until 40┬░C. The protease produced from WI isolate might be able to be used in the processing of fish sauce.
