Determination of endothelial cell density of corneal transplant by digital image processing method

The cornea is the transparent front part of the eye which provides most of an eye's optical power. Several indications caused the cornea to be replaced by transplantation. Penetrating keratoplasty have so far been the most successful of all human organs transplantation. However, endothelial density level of 2,300-3,300cells/mm2 is a compulsory condition to ensure a successful transplantation. This thesis aims to develop simple software which is able to determine the overall endothelial cell density of a corneal transplant as an alternative aid for ophthalmologists. Digital cell image is acquired from a specular microscope from Indonesian Eye Bank. Basic algorithm of digital image processing such as thresholding, histogram equalization, erosion, opening and labeling are used in MATLAB. Cell density result count of 1987 cells/mm2 shows 80% of accuracy compared to 2488 cells/mm2 obtained from commercial software. Thus, developments of more advanced algorithm are welcomed in the future for better outcome.
