Identification of Carotene and Beta-Cryptoxanthine in Crude Palm (CPO) Sludge Under Flow

Due to huge number of crude palm oil (CPO) plantation and production in Indonesia, which automatically followed by the amount of its sludge production, the research was conducted to identify carotene and beta-cryptoxanthin in the CPO sludge under flow and to find new source of carotene and/ or beta-cryptoxanthin for food industry. the CPO sludge under flow is extracted using choloroform to separate the carotene and beta-cryptoxanhin from other compounds. It is put in room temperature until turns into dry remnant for quantitative determination of carotene using a spectrophotometer. There were two treatment factors, ratio of CPO sludge under flow and chloroform (50:50, 60:40 and 70:30) and mixing time (5 minutes and 10 minutes). the acquired data is later being analyzed using anova two-factor with replication followed by post-hoc analysis (Tukeys HSD) to determine the best treatment sample. the best treatment sample was then analyzed using HPLC for the identification of beta-cryptoxanthin. There was carotene detected in CPO sludge under flow, but beta-cryptoxanthin was not detected. Both treatment factors significantly affected the carotene content in CPO sludge under flow, for shorter mixing time (5 minutes) overall has more carotene content in CPO rather than longer mixing time (10 minutes). the ratio 60:40 of CPO sludge under flow and chloroform had the most carotene content in CPO compared to other ratios at 5 minutes mixing time, while the ratio 70:30 of CPO sludge under flow and chloroform had the most carotene content in CPO compared to other ratios at 10 minutes mixing time.
