Preliminary study of antivirus for human papiloma virus (hpv) using protease enzyme bromelaine
HPV is a virus that causes cervical cancer. The nature of the HPV morphology makes it a prime specimen to be treated with the protease Bromelaine. This preliminary research towards the effect of Bromelaine towards HPV was done in several steps. This includes sample collection, sample analysis, treatment of the sample by Bromelaine, analysis of the DNA concentration of the treated sample and finally by using PCR and gel electophoresis . To analyze the difference between treated and untreated sample, control samples were used which were only mixed with PBS buffer. The treatments of the samples using Bromelaine U which has specific activity of 5.9/mg protein, volume activity of 150.9 U/ml, and incubation time of 6 hours at 37oC, show that the DNA concentration of the samples increased by 164.8 % and 270.8%. Visualization of the treated samples using the results of PCR by way of gel electrophoresis confirms that the viral DNA in the treated samples rised which were displayed as much more prominent and clear bands in the agarose gel.
