Determination of atpase in human blood as an indicator in overweight and underweight people

ATPase is a membrane-bound enzyme that hydrolyzes ATP to ADP, produces free energy to pump ions across cell membrane. This study hypothesized the possibility of erythrocyte membrane total ATPase activity correlation with to human obesity determined by BMI. However, until present, no research correlates obesity to total ATPase activity. This study attempted to determine a method to measure optimum total ATPase activity and determine the correlation between total ATPase activity and obesity determined by BMI. Methodology used in the assay is based on the oxidation rate of NADH correlates to ATP hydrolysis. The optimized assay reagent and blood processing method determined in this research is optimal to measure erythrocytes membrane total ATPase activity. Study that involved 9 male subjects with average age 22.56 with BMI range from 17 to 36 showed that there is no correlation between total ATPase activity with BMI. The average total ATPase activity obtained from these subjects at 25┬░C is 0.0029 ┬▒ 0.0011 Unit/mg proteins.
