Purification of -Amylase Enzyme Produced in Solid State Fermentation Using Sorghum Grain as Substrate

Amylases are one of the most important and oldest industrial enzymes. Alpha amylase involve in energy production by breaking down starch into sugar that is used later to produce energy. Sorghum grain can be used as a substrate in the production of ?í¬±-amylase in Solid State Fermentation. in purification of ?í¬±-amylase in Solid State Fermentation using sorghum grain as substrate, multi-precipitation and drying technique was adopted. Every stages of precipitation at pH 5, pH 6, pH 8 and pH 9 were analysed to obtain the specific activity of enzyme using reducing sugar by DNS method and protein content determination. Multi-precipitation process of enzyme by pH shifting was capable to increase specific activity enzyme using the multi-precipitation process followed by drying in purification increased the specific activity of the enzyme by 41.5X U/mg.
