Purification of high grade bromelain from pineapple by using ion-exchange chromatography technique

Most people do not concern that pineapple also contains very valuable natural enzyme which is proteolytic bromelain enzyme. Bromelain is believed to have many benefits and is very promising to the development of food industry, as well as medical purposes. The purpose of this study is to isolate and purify bromelain enzyme, in order to yield higher grade products. The bromelain extraction method using pineapple?s stem and fruit in this study was done in several consecutive processes. These included the separation of inner cell components by centrifugation, purification by size exclusion chromatography using Superfine Sephadex G-50, addition of ion-exchange chromatography technique on CM (Carboxy Methyl) Sepharose fast flow, and followed by freeze drying as the final step. Each treatment was found to produce different effects and yields on the purification level. The purification of high grade bromelain in this study yielded 0.162% bromelain in pineapple stem with total protein content of 18.299% and total specific activity of 17.645 U/mg protein. Whereas, pineapple fruit yielded 0.138% bromelain with total protein content of 21.340% and total specific activity of 11.079 U/mg protein. Stem bromelain has been proven to have higher enzymatic activity than fruit bromelain.
