Purification of high grade bromelain from pineapple
Pineapples contain bromelain, a phytochemical compound that was proven to possess a lot of benefits. These proteolytic enzymes are likely to be very promising to the development of food industry, as well as medical uses. The purpose of this study was to isolate and to purify bromelain enzyme, in order to yield high grade products. The bromelain extraction method using stem and fruit of pineapples in this study was done in several consecutive processes. These were including the separation of inner cell components by centrifugation and purification by size exclusion chromatography using Superfine Sephadex G-50, then followed by freeze drying. Each treatment was found to promote different effects and yields on the purification level. The functional assays were also performed, including protein content test by using Lowry method and enzyme activity test using casein as subtrate. Stem bromelain had rendement of 0.176% with total protein content of 19.180% and enzyme activity of 16.099 U/mg protein. Whereas, fruit bromelain had rendement of 0.163% with total protein content of 22.255% and enzyme activity of 10.723 U/mg protein. Stem bromelain has been proven to have higher activity than fruit bromelain.
