The study of desalting techniques on chitooligosaccharide production
The aim of this study was to remove salt from chitooligosaccharide with three desalting technique and determine the best desalting technique. The desalting technique used were ultrafiltration with hollow fiber cartridge 5000 NMWC, dialysis tube with 10000 NMWC and gel filtration chromatography containing HiPrep 26/10 desalting with G-25 superfine sephadex as stationary phase in the column. In order to detect the presence of chitooligosaccharide, TLC method was used, followed by MALDI ? TOF ? MS to detect low concentration of chitooligosaccharide. Qualitative analysis of salt presence was identified using silver nitrate method, while the quantitative analysis was done with Volhard?s method. Ash content was measured using ash content method. Results showed that ultrafiltration could not remove most of the salt. While gel filtration chromatography and dialysis, were able to separate most of the salt from chitooligosaccharide. However, the best desalting technique was gel filtration chromatography with HiPrep 26/10 desalting because it has more complete chitooligosaccharide with various degree of polymerization occurred in the result.
